2025 Inborn errors of immunity practice parameter

The current US practice parameter on inborn errors of immunity (the term now used in place of primary immunodeficiency), issued jointly by the Joint Task Force on Practice Parameters with AAAAI, ACAAI and the Clinical Immunology Society. It reflects developments since the 2015 parameter, including genetic testing for diagnosis and treatment selection, newborn screening for severe T-cell lymphopenia, and targeted or precision therapies, and covers management topics such as immunoglobulin replacement, antibiotic prophylaxis, haematopoietic stem cell transplantation and quality-of-life assessment.

    The 2025 parameter abandons the previous edition's structure of working through individual genetic entities and instead organises recommendations into a diagnosis part and a management part. Diagnosis now covers newborn screening for severe combined immunodeficiency, genetic testing and comorbidity surveillance, while management addresses immunoglobulin replacement, antibiotic prophylaxis, haematopoietic stem cell transplantation, precision medicine and quality-of-life assessment.
